Research, Innovation & Impact (RII) invites applications for an Assistant Staff Scientist for its Analytical and Biological Mass Spectrometry (ABMS) Core. The Core provides a variety of mass spectrometry services in a well-equipped University-wide facility. The successful candidate will be primarily responsible for providing high-level qualitative and quantitative proteomics analyses to both University of Arizona and external researchers. Assays will include label-free (PRM, DIA) or labeling techniques (iTRAQ, TMT, SILAC, or NeuCode) on Orbitrap Exploris 480, Q-Exactive Plus instruments, and an ALTIS triple-quadrupole mass spectrometer. The successful candidate will be involved in consulting with researchers, participation in experimental design, sample preparation, LC-MS/MS analysis, data analysis interpretation, and instrument management. Related responsibilities will include optimization of assays, development of new analytical assays, and monitoring scientific literature. The candidate will be expected to assist the Manager in the day-to-day operation of the Core. This position will report to the Manager of the facility. Duties & Responsibilities:
Receives direction from the Manager to address major research goals of the lab. Uses knowledge and technical expertise in sample preparation, separation, and analysis by mass spectrometry and data processing to complete research projects. Prepare samples using various standard in-solution and in-gel proteomic sample preparation protocols for bottom-up mass spec data collection as well as with alternative workflows, such as FASP, S-trap, etc. Development of MS-based methods for scientific projects and acquisition, evaluation and analysis of data (label-free (DDA, DIA, iBAQ) & label-based (SILAC, TMT, iTARQ) quantification, targeted proteomics (SRM, PRM), PTM/Phosphoproteomics, low-input proteomics, disulfide linkage, and Top-down analyses. Proteomic data processing with vender specific proteomic data analysis software platforms (Proteome Discoverer), as well as third-party software platforms such as MASCOT, Max Quant, Byonic, Skyline, and Scaffold. Knowledge and experience with programming platforms such as R (R Studio) and other informatics tools such as MS Stats is a plus. Intact protein analysis by mass spectrometry. Support and maintenance of LC-MS instruments, routine performance monitoring (calibration, QC measurements), including instrument troubleshooting and front-end cleaning. Prepare samples and perform nano-LC-MS/MS analyses using QExacitve Plus, Orbitrap Exploris 480, and an ALTIS triple-quadrupole mass spectrometers. Excellent written and verbal data presentation skills and the ability to use software tools to produce high-impact presentations of data, highlighting key takeaways. Detail-oriented with the ability to capture and record complex scientific data accurately and reliably. Attends weekly team meetings to discuss results, plan experiments to ensure that projects support current team goals in a dynamic environment. Assist in the operation and routine maintenance of analytical instrumentation including other mass spectrometers in the facility. Additional duties may be assigned.
Knowledge, Skills & Abilities:
Excellent interpersonal and oral/written skills to interact with a diverse group of graduate and undergraduate students, staff, faculty, and visitors. Ability to understand and perform chemical derivatizations on samples to be analyzed. Basic understanding of the theory and practice of mass spectrometry. Strong computer skills with the concepts and practices of the field of Bioinformatics as it relates to mass spectrometry and experience with MS-based Omics data analysis systems and software. Knowledge of MS software (Thermo, Bruker), database search and evaluation software (MASCOT, MaxQuant, Perseus, Proteome Discoverer, PEAKS, Spectronaut, Scaffold), and associated bioinformatics tools (R, Python) is advantageous. Open-minded and motivated personality with a focus on team play. Knowledge of MS software and evaluation software (Proteome Discoverer, PEAKS, MASCOT, Scaffold) and associated bioinformatics tools (R, Python) is advantageous. Excellent knowledge of written and spoken English.
Minimum Qualifications:
Bachelor's degree or equivalent advanced learning attained through professional level experience required. Minimum of 5 years of relevant work experience is required.
Preferred Qualifications:
Ph.D. or equivalent degree in a field related to mass spectrometry or analytical sciences and 1-2 years of related research experience; OR Masters' degree and 5 or more years of related research experience; OR Bachelors of Science and 8 or more years related research experience. Experience in sample preparation and using nano-HPLC-MS/MS for proteomics analysis is preferred. Experience in proteomics data interpretation is preferred. Comprehensive, hands-on experience using mass spectrometers and nano-H/UHPLC systems is mandatory; experience using Thermo instruments is advantageous. Experience in the development of MS-based methods for proteomics projects and acquisition, evaluation, and analysis of data (label-free (DDA, DIA, iBAQ) & label-based (SILAC, TMT, iTRAQ) quantification, targeted proteomics (SRM, PRM), PTM/Phosphoproteomics. Experience in R programming is a plus.
